Inhibition of IL-2 Dependent CTLL-2 Proliferation by Immune Complex from Patient with Ovarian Cancer

Abstract
A variety of patients exhibit defects in cell-mediated immunity, particularly cancer patients are one of the most severely immunocompromised, but the precise mechanisms remain unclear. A number of serum factors such as immune complex, fetal
haptoglobin
variant, fibrinogen, and other acute phase reactants are known to be increased in parallel to tumor progression in cancer bearing patients. And these non-specific immunosuppressive factors may suppress cell mediated immunity of the host
profoundly.
Considerable number of evidences are indicating that major components of cell-mediated immunity against various types of solid tumors are cytotoxic T cell(Tc), NK cell and K cell of the host. Among these three Tc is known to be the most potent
effector
component, Based on these concepts we studied immunosuppressive potential of ascitic from 9 ovarian cancer patients and immune complexes extracted from it in the murine cytotoxic T cell(CTLL-2) model by testing inhibition activities of ascitic
fluid and
extracted immune complex on the IL-2 dependent CTLL-2 proliferation. It ws shown that nonspecific anti-proliferative activity of cancer ascitic fluid was increased in a dose-dependent manner. And this activity could be precipitable by saturating
with
33% ammonium sulfate solution. It turned out that imune complex in this 33% ammonium sulfate-precipitable protein fraction was the most prtent suppressor on IL-2 dependent CTLL-2 proliferation.
This result suggests that immune complex in ascitic fluids from patients with ovarian cancor suppress the immunoregulatory network especially by inhibiting antitumor cell mediated immunity and may lead to better understanding of immunosuppression
and
development of cancer cells despite of immune surveillance.
